How popular do you need to be before setting up a patreon/subscribestar and expect it to be healthy? (Around 300-500€ at an absolute minimum)

Depends on your art and what you offer. There are some big accounts that don't really make much money from Patreon and smaller ones that actually make a bunch since they offer a lot on Patreon or just actually advartise their shit better.

It varies a lot, but a conservative estimation is a 1% conversion from your marketing funnel (i.e. twitter).
So if you change 5€ per month, with your target of around 500€, expect to need at least 10k followers.

That's not how you approach it. Open a Patreon now and grow it gradually. It doesn't really matter how popular you are. Money isn't your goal right now.
You can use Patreon as just another place to share your work. Only NSFW content needs to be behind a paywall for age verification. Everything else can be free and people can follow you without paying.
Focus on getting non-paying Patrons first. Without non-paying Patrons nobody will see your paywalled posts. Patreon will promote paywalled posts to your non-paying Patrons who may then decide to upgrade.

Character polls are fine but don't let individual followers suggest things in front of everyone else. That's how drama starts. You will inevitably get a few really really annoying followers.
The vast majority of your paying followers will never say a word. They will however quietly judge you from the shadows.
WIPs are great conversation starters in the group chat and you can turn access to that chat into a reward tier.
