Some of it you replace quite quickly - you make up the fluid volume in a day or so - but the red and white cells, and (to a lesser extent) some of the other stuff, take longer to manufacture. As a one-off, you can stand to lose that much; but if they kept taking blood out of you every two weeks, after a couple of months you'd be severely anaemic and immunocompromised, and your blood wouldn't clot properly, and a number of other things too.
