People figured out decades ago that almost all people fidget, sweat and get nervous when they know they're lying (espcially if the stakes are high, i.e. prison time). Then some other people figured out how to make a machine that detects if you're doing too much of those things while you're being questioned. Then some other people standardised this process and called it a polygraph test.

The main way the polygraph test work and the only way it works is when the interrogated person has both something to hide and something to gain when their lying. It's not impossible to lie, the general thing is to not give a fuck. You can watch the mythbusters episode about it, Grant manage to fake it. You need a lot of self conviction/self hypnose to fake it.
