It was in the room with the giant nest after fighting the Plagas armor dudes, which is also a pretty hard fight (especially on professional) but you can cheese it with flash grenades after the parasite bursts out of their heads

I've done two story runs on professional difficulty already. The first time around it can get pretty brutal, especially in the castle where enemies swarm the fuck out of you and Ashley dies in two hits and you die in like 2-3 hits.

The castle section also forces you to fight in some tight ass spaces, probably the hardest section of the game IMO

The hardest segment is arguably the statue room where ashley is on an upper level but the semi automatic trivializes the game
issue is on professional the crossbow men have max aggression
