Is the location if Ausport Castle, on the small island in the southeastern corner, sensible? I thought it might be useful for maintaining control of the island and straight and could provide support to the nearby coastal towns if necessary, but am not sure if it would make more sense in the mainland.

Why wouldn't it? It's a large enough island that it could serve as a staging point for raids or invasion. If there is a port city of any importance in that straight that island would have a garrison of some sort. If you are uncomfortable with it being a huge ass castle or fortress just make it a small wood fort or a tower with a wall.

I guess pirates/foreign raiders/other dastardly types may have have had a habit of setting up base camp on the island, and so a castle there to lock it down might have been considered useful? Hell, the castle may even have been (in part or in full) constructed by said villains, and taken from them.

The main island must be fuckhuge if it has several biomes. That smaller island is still easily big enough to host a major settlement.

Ruling the island from the mainland would be inconvenient. If its large enough to sustain a permanent population there will be some guy in charge, and he would want a good place to rule the island from.

I'd say it'd be stupid to not have the island next to a major city fortified, especially if naval warfare is a realistic expectation. It's a fast response to any unrest in the mainland city and nearby areas, it makes attacking by sea very difficult because it'll naturally pincer any invasion on ships. Attacking the fortress isn't easy either with the city able to respond in kind to any assault.
You can throw in some historical event regarding the fort in the background to subtly reinforce the idea ofc, look into similar places irl. Suomenlinna could be something, visited there recently on vacation.

Might be to do with geography. If the nearby city is in a valley/delta due to the river, it may not have a convenient place to build a castle foundation. If the island has a good bedrock to work with and a source of freshwater well then I guess the gods want a castle on it.

The land was indeed formed by Gods and their will should be taken into account as they can be mercurial. I’m thinking with natives on the island and threats from the wilderness the location works. I had mostly been concerned about the separation across water being a hindrance rather than a help and if it were even needed.
