I feel the biggest thing pointing towards Mari being the true love is that (other than with the prince) the only three times the fish dagger has reacted have been in relation to her. First when Mari tells Lara she can't stay in their house forever, the feeling of rejection makes the dagger briefly show a reaction that looks like it's thinking about eating Mari's soul. Second time is the big blade spawning when Mari says she likes Lara. Third time the blade moved to attack Kota when Lara thought Mari was in danger. It has to mean something that they make it react only to her.
