Is it possible for an autistic man to develop his social skills through practice and great effort such that he becomes suave and good with the ladies?

I don't know about becoming good with the ladies, but yes. I was a NEET almost all my life until 5 years ago. my first job was extremely rough because i didn't know how to socialize and would creep people out. fast forward to my 4th and 5th job, I can hold a conversation quite well, and become chummy with people who want to talk. i'm still introverted and largely avoid others, but anyone who wants to talk, i can chat them up and charm them.

socializing is a skill like any other and can be honed with practice. get a shitty part time job and interact with your coworkers. go to community college and take a few easy classes. you'll build social skills easily.

Yes. I've mostly pulled it off. You don't even need to develop social skills. 'Fake it till you make it' long enough and you eventually become the person you want to be.
