You'd have to go back to the early days of cartoons for that. Say, Pete from the early Mickey Mouse shorts, Bluto from Fleisher's Popeye, or any of the mad scientist villains from the old Superman cartoons.

Off-hand, I'd say the first character to _define_ "cartoonishly evil" wasn't even a cartoon, it was the Wicked Witch of the West from The Wizard of Oz. Margaret Hamilton's performance was so over-the-top it set the bar for all future movie villains, animated or live-action.
