In the Marvel universe, space colonization would involve possibly treading on the property of hundreds, if not thousands, of aliens which are significantly more advanced and equipped than you. It's that those types of developments *can't* happen because the universe has to be tied to the status quo of the real world as a convention of the genre. It's the same reason superheroes are always fighting against injustice but never seem to make any meaningful social change. If they did the story would be over or the world would deviate too far from our own to be relatable to the audience.
