What do you mean "kicks in". Taking exogenous estrogen and an anti androgen will put you in cisF levels within a week. Effects such as skin softening and breast development will restart on the same timeline as if you were starting estrogen for the first time. Hormones aren't cumulative, they just switch on and off certain body processes
