Fluttershy and Pinkie Pie are most peoples go to choices, but I think Rarity and Applejack are pretty underrated. They both have carnivorous pets, meaning they'd be more sympathetic to a human's dietary needs, and while Rarity might be a tad racist at times, neither of them are ever really particularly intimidated by none pony creatures.The real hurdle would really be body language. Without pony ears and tails and just the other natural behavioral differences, they could have a harder time understanding human social cues. So basically worst case scenario is that you have a really bad case of resting bitch face which makes you seem a bit unapproachable.

Rarity, Fluttershy, etc) vs ones who don't and so the eyes are something that disturbs them even if they may not be able to fully realize what exactly is creeping them out. To ponies not used to it, basically any prolonged stare would feel like you're glaring at them. Off topic, but early Kengan Asura chapters I think illustrate a bit what it would be like, where the characters are purposefully drawn to look a bit creepy and hostile even when their just in a casual situation. Ponies not used to being around predators I imagine would have a perception closer to this.

As a result to ponies it would seem like you just stare really, really uncomfortably hard at anyone you speak to.The important part is that this effect wears off the more familiar the pony becomes with a human. If the mane 6 or similar are close friends with them, then by that point they should know you well enough to be able to discern human body language. There would be a learning curve to it, but once ponies are accustomed to how your body language is different from theirs, they wouldn't find you so weird looking anymore.
