You could also pretty easily trick her into hurting herself I think. Her death would also affect ponyville the most which would add to Twilight's anger. Fluttershy is not trusting but she is probably the easiest to take out "conventionally". Her innocence would also make Twilight extra angry because she would have difficulty processing how anyone could be so cruel to a pony like Fluttershy. This probably applies to Pinkie as well though. Applejack would probably be the most difficult followed by Rarity and Dash.

And when she was ready, make her the ruler of Equestria. Even worse is that both Celestia and Luna retired at the same time, so now basically Twilight has to take care about both the moon and the sun.

This is why it would've made much more sense to at least have Starlight become an alicorn too to keep her company and most importantly sane. Celestia had Luna and even during the 1000 year absence she had hope Luna would eventually return. Once Twilight's friends dies that's it, all that's left is resurrection magic which if Full Metal Alchemist is anything to go by never turns out well. Twilight turning evil and having to be put down to rest by a new m6 would be a pretty metal way to start G5.
