Combining material slots seems to be one of the most effective things you can do beyond baking in your non animated blendshapes. As for shaders, poiyomi can add quite a bit of tax if you start animating multiple properties at once but if you really want to be optimal then using the mobile shaders would be best. Warren's fast fur shader looks lovely but just doesn't seem to run very due to how Z-Buffers work when closer to the viewport. Can't say I've tried mochie or liltoon enough to have an option on them.

A single file can almost always be compressed more efficiently which is effectively what your vrca file is acting as over a group of files. I'm sure someone else can provide a better answer though, I too am a total retard.

every unique material is a separate draw call and lots of draw calls are bad for performance. you can combine materials but it requires uv work and baking mats together which is work and makes it harder to make changes

quest forces you to use built in shaders so they can be batched more easily. when people say the devs "need to optimize the game" that's what they're actually asking for without realizing it. you can either have the flexibility of custom shaders or the performance of standardized shaders but not both

d4rkAvatarOptimizer's tool seems pretty good at doing the heavy lifting for you, It's been quite a nice save to not have to edit uvmaps like back in the SL days of things.

every material creates a submesh and adds a draw call. a single mesh w/ three different materials is effectively three meshes.
more meshes makes it more complex for the CPU and means the GPU does multiple passes once the CPU sends it along. it is cheaper to render one complex thing (one pass) than three separate things that add up to the same complexity (three passes).

Also regarding this whole performance thing, complex animation controllers and physbones is a silent killer, an extra material like, piercings gets evaluated fast despite being an extra drawcall since all the unused shader features get treeshaked anyway, most of the time your cpu is choking on the former two since it's all done on the main thread and is too big to stay cache resident.

This is why I bothered to go the extra mile and use root bones to combine physbone scripts into children bones instead. No need for two separate ear physbones, combined the belly and breasts as well.

Also while I'm at it, I've found direct blendtrees to be far more optimal than normal state driven behavior, recommend trying that out if you can.
