It's because of the delivery, the joke itself, and the reaction to it from the other guy. All of it is funny. His partner tells him to act like a retard and he comes up with the actual perfect thing to do by doing the downie voice. Then the other gangster's reaction was funny because he was like, "alright cool"
You wouldn't expect a gangster to just be okay with a retarded guy, you'd expect him to make some kind of joke about it or make fun of him, but he doesn't, which is funnier
