He had a cartoonish personality because he's Steve Ditko's not so subtle jab at Stan Lee and his hatred for the media (which is pretty based). They tried to retcon him into being more of a good guy later on but by that point his shtick was already established

Spider-man can only exist as he does in a fantasy universe where most people are willing to believe in the simple goodness of superheroes while men like Jonah are a grumpy minority. That has changed over the years. Sometimes he's upset that Spider-Man upstages his son, other times he genuinely does consider him a threat/menace, but he pretty much always has hated Spider-Man since his inception in the 60s to now.Jameson has spent a silly amount of money trying to get Spidey killed or captured. He once became mayor and also created a special Spidey task force he would regularly pay overtime (a big deal since he's Mr. Krabbs levels of greedy) to try and hunt him down.

He's a minor character at first who exists more as Peter's boss (and Betty Brant's boss since they date) who sometimes sends him on assignments and is a money grubbing cheapskate. When I was a kid I had a marvel poster that showed NYC with the different territories of heroes marked out along with various points of interest.
I haven't read much of the comics, but I'm guessing that at some point they had a story where JJ discovered Spidey's identity, so that readers could see his reaction, but then his memory of it was erased before the arc was over and all went back to normal? And there's probably some stupid multiverse story where Jonah is Spider-Man and Peter is the editor.
