I would Just clone Devil May Cry

Make it where you play as Seras or Walter and just have Alucard as an unlockable once you beat the game. He'll be unbeatable and you can steamroll the game as a bonus mode.
Or just make it so his "health" is tied to the number of souls he has/can collect. If he takes damage, he loses souls and could possibly lose some abilities or powers. As he gets weaker he gets slower and shittier, enforcing the idea that while you can't die, you don't want to take damage either.
Or do a risk reward system where yes, Alucard can "die" but if you time counter's properly, you can absorb all the incoming damage and turn it back on the enemy as an attack/taunt like how he does in the anime. Purposely letting himself be destroyed just so he can come back and scare the shit out of people before eating them.

Base it on the older 13 episode anime, the parts before Incognito. Play as human members of the Royal Order who go out on various vampire jobs around UK and Ireland. Play a bit into their competition with the Roman Catholic church.
