Set up your city, set up its major players, possibly introduce your players to the setting and the basics of unlife if they're completely new to things, but then give them a good degree of autonomy. Vampire is at its best when you have at least one PC with ambition and goals of their own, because just doing "quests" for older vamps is a dead end. Good when you're a youngster looking for experience, cash, and favors from more powerful vampires, but not something to spend forever doing. Make it clear that they're not the destined heroes, and that the world will move without them. Have schemes running in the background by various NPCs, and have those plots move organically around the PCs own plots. This is the unfortunate truth about VtM, it's focused a ton on characters rather than having to save the world or win the day, so the PCs do more heavy lifting for the game than usual. Putting the impetus on them to do things, or else have unlife and its opportunities pass them by not only stirs them into action, it also helps the feeling that the world does not revolve around them. It encourages their active participation in the Jyhad, despite all the danger and misery held within. Oh, and never skimp on the mortals. It shouldn't always be just vampire politics. Humanity (though in my opinion it's a deeply flawed and honestly crummy system mechanically) is important, not just the humanity within, but the humanity without.

One of the best TTRPG experiences I ever had was climbing the ladder of Clan and Camarilla as a 12th generation Ventrue. Another piece of advice. Humanize your vampires, but don't make them dindus. Some will be better than others, but nobody's ever a pure cartoon villain, and nobody's ever without sin. Humanizing traits don't even need to be positive, just... human.
