Wadjet Eye's probably the biggest modern point and click developer but I rarely hear anyone talk about their games (not a knock on their quality, just that it's hard for point and click adventures to get noticed anymore). We'll probably never get a big mainstream point and click ever again especially since the last serious attempt was Broken Age.
