A collector bought a proto version of Punch Out in an auction for $45,000 and allowed the video game history foundation to dump the ROM. It turned out to be identical to retail except for a couple of bytes. No refunds I guess.

The prototype in particular isn't drastically different from the final, but the differences found so far are pretty significant from a development standpoint (most notably Pizza Pasta and Piston Hurricane were meant to be in the game alongside a couple of unknown names). It's incredibly telling that this thread is in bad faith considering this is a best case scenario for a prototype cart auction.

So Summoning Salt removed the video, it had a lot of differences like unfinished move sets and placeholder fighters that just cycled animations. Not really much unseen content which is the most exciting part of protos.

Printed label, (IMO this is the biggest indicator, why would this ever have a printed label, even if it was intended as black box, why would they comm different art for a proto that still is full of placeholder shit?)
Text not matching up with known proto screenshots further along in dev (the fonts, along with different heart values, though this COULD be chalked up to how it was captured, and debugging for the different health amount, but IMO the differences especially in the 0's make it clear.)
Cifaldi himself states that it is engineered unlike any prototype he's ever seen chip wise in the video he has up. He couldn't even declaratively say it was real leading up to it.

A few things.
A game can change something, then revert it to a previous state. Just like how something might work in one build, break in a later one, then be fixed for the final.
As for the bizarre PCB, the board itself would have been a prototype. Punch Out uses a unique mapper that was used by no other game, so they were probably in the process of prototyping the PCB itself. As for the apparent usage of mask ROMs as opposed to EPROMs, who knows. There'a a good chance there were once several more copies of this, and they were either destroyed, lost, or just haven't turned up with.
As for the label, it's just a mockup, either to make it more presentable for whomever they were demonstrating it to, for marketing photos, for experimentation, or because some intern wanted to show initiative and decided to print out their current label mockups and slap them on their prototypes.
