Is 250 gto a predecessor of 288 gto and therefore one of the flagships or a completely different car?

No.

Its a Aston Martin DBR9 GT1 equivalent to an Aston Martin Valkyrie, which is to say they have fuck all to do with one another.

250 GTO wasn't a flagship, it was literally just a race car.
by comparison 288 GTO was only made to sell to customers to make money. so definitely not the same type of car.
you could look at it like the E30 M3, which was a racing homologation car, compared to every M car after that being just a road car.

The 288 is a joke, the 250 was a very successful racer. The 275 GTB Competizione was the (faster) successor to the 250 and a bargain today at only $10,000,000.
