It always had co-ed marketing. The reason it got greenlit was because of the proven the co-ed appeal of Smallville. Smallville was comicshit to appeal to guys, plus hot guys to appeal to girls. Supernatural was horror to appeal to guys, plus hot guys to appeal to girls. It was somewhat similar to the marketing of Charmed: girls like witches, plus some cheesecake of the actresses to appeal to guys.
After season 5, Supernatural decided that their audience was women which is why the new show runner was a woman. I think that late 90's early 00's horror boom was over and the 'fantasy for girls' with Twilight and Vampire Diaries was picking up so they decided to pivot to that.
