We have no way of knowing whether all the Great Runes we collect are comprehensive of what the original set looked like or if some that would complete the set are gone. Or even whether everyone that has a rune in the present tense had one in the past tense. Someone like Godrick could have easily gotten a Great Rune late in the timeline after Vyke was lost to Frenzy and then gave up his Runes.

You've got 2 days (roughly 10-15 min each) of exploring shifting points of interest and killing bosses to get runes, weapons, buffs, etc. At the end of each night, you fight a boss. The third day is entirely dedicated to fighting the Nightlord. Each character has a special moveset with certain weapons and some unique skills. If you die, your teammates can revive you, but it gets harder each time you go down until you hit a site of grace.It's genuinely fun and is a great way to try out weapons and builds you wouldn't normally bother with in the base game.

Since Vyke is the one publicly considered to have gotten the closest to being fit for lordship, it was probably Vyke. We have no clue where his Great Runes came from. The best guesses with the demi-gods we know weren't murdered in the Night of the Black Knives are Godefroy and Ranni. In each case, Vyke would have had to have basically been handed Godefroy's Great Rune after Kristoph defeated Godefroy (or got the finishing blow), and he'd have either picked up the shards of Ranni's Great Rune if she destroyed it or otherwise found it if she discarded it intact. As for where his Great Runes even went, maybe the Three Fingers burned them out of existence or something, a la what seems to happen to the Elden Ring in the Frenzied Flame ending. Of course, the Three Fingers doesn't do that to the main character, but you could totally brick your playthrough if it did with no means to repair the Elden Ring. You would have to hope a single mending rune sufficed, get at least two mending runes (you can also brick yourself out of this), or choose the Frenzied Flame ending (which you could then brick yourself out of by purging the Frenzied Flame).Also, in the game itself, it's pretty clear that the Frenzied Flame made Vyke go full schizo and he murdered his own Finger Maiden, the person he was trying to save. The Frenzied Flame is an asshole.

But what it also means is that he was gone for potentially thousands of years after the Tarnished that would theoretically have a place of honor in being descendants of his followers, started reappearing. Morgott still remembers that Tarnished are meant to have that Godfrey-follower warrior blood in them, but by this point he mostly just hates the Tarnished for being by nature power hungry and causing more instability. Others might not even remember the first part.
